Protection was independent of exosomes under the conditions tested. In mice, injecting cMSC-conditioned media into the infarct border zone reduced apoptotic cardiomyocytes > 70% locally. Thus, hPSC-CMs provide an auspicious, relevant human platform to investigate extracellular signals for cardiac muscle survival, substantiating human cardioprotection by cMSCs, and suggesting the cMSC secretome or its components as potential cell-free therapeutic products.Successful execution of daily activities requires accurate perception of the torques one generates about multiple joints. Even so, previous studies are mostly limited to an individual’s perception when torques are generated about a single joint. Consequently, this study investigates how accurately individuals judge torques at their arm during a multi-joint task. Puromycinaminonucleoside The accuracy of fifteen right-hand dominant participants (age 60 ± 10 years) in matching isometric elbow torques, within the same arm, was quantified during single- and/or multi-joint tasks. Participants generated and matched elbow torques when the shoulder was (1) not abducted (single-to-single-joint), (2) abducted (multi-to-multi-joint), and (3) abducted and then not abducted (multi-to-single-joint). The constant error for the multi-to-single-joint condition (dominant 6.9 ± 5.9 Nm, non-dominant 6.0 ± 5.5 Nm) was greater than that for the single-to-single-joint condition (dominant 2.7 ± 3.1 Nm, non-dominant 3.4 ± 2.8 Nm) (p less then 0.001) and multi-to-multi-joint condition (dominant 3.0 ± 2.8 Nm, non-dominant 3.9 ± 2.7 Nm) (p less then 0.001). The constant error for the multi-to-multi-joint condition did not significantly differ from that of the single-to-single-joint condition (p [Formula see text] 0.780). Findings indicate that in older adults the perception of a self-generated torque during a 2-degree-of-freedom (DOF), multi-joint task is largely influenced by the motor commands associated with the 2-DOF task and is not specific to the DOF at each joint.Stable and flexible super-hydrophilic nanotubular-based titanium oxide electrode has been utilized as the active electrode of self-powered humidity sensor. TiO2 nanotubular electrodes fabricated through anodization method and utilized in combination with Kapton electrode as the triboelectric nanogenerator (TENG). Vertical contact-separation mode TENG performance has been examined in various range of frequencies and the maximum output voltage and current more than 300 V and 40 μA respectively with maximum power of 1.25 ± 0.67 mW has been achieved at 4 Hz. The fabricated TENG has been employed as the active self-powered humidity sensor. Super-hydrophilic feature of TiO2 nanotubes resulted in full absorption of water molecules, and noticeable decrease in charge transfer across two triboelectric materials upon increasing humidity. The TiO2-based TENG sensor was exposed to various relative humidity (RH) and the results showed that by increasing the humidity the output voltage and output current decreased from 162.24 ± 35.99 V and 20.4 ± 4.93 μA at RH = 20% to 37.92 ± 1.54 V at RH = 79% and 40.87 88 6.88 ± 1.7 μA at RH = 84%, respectively, Which shows the responsivity more than 300%. This method of measuring humidity has a simple and cost-effective fabrication that has various applications in many fields such as industry and medicine.Exercise-induced pulmonary hemorrhage (EIPH) is a common condition in sport horses with negative impact on performance. Cytology of bronchoalveolar lavage fluid by use of a scoring system is considered the most sensitive diagnostic method. Macrophages are classified depending on the degree of cytoplasmic hemosiderin content. The current gold standard is manual grading, which is however monotonous and time-consuming. We evaluated state-of-the-art deep learning-based methods for single cell macrophage classification and compared them against the performance of nine cytology experts and evaluated inter- and intra-observer variability. Additionally, we evaluated object detection methods on a novel data set of 17 completely annotated cytology whole slide images (WSI) containing 78,047 hemosiderophages. Our deep learning-based approach reached a concordance of 0.85, partially exceeding human expert concordance (0.68 to 0.86, mean of 0.73, SD of 0.04). Intra-observer variability was high (0.68 to 0.88) and inter-observer concordance was moderate (Fleiss’ kappa = 0.67). Our object detection approach has a mean average precision of 0.66 over the five classes from the whole slide gigapixel image and a computation time of below two minutes. To mitigate the high inter- and intra-rater variability, we propose our automated object detection pipeline, enabling accurate, reproducible and quick EIPH scoring in WSI.The mechanisms mediating suppression of reproduction in response to decreased nutrient availability remain undefined, with studies suggesting regulation occurs within the hypothalamus, pituitary, or gonads. By manipulating glucose utilization and GLUT1 expression in a pituitary gonadotrope cell model and in primary gonadotropes, we show GLUT1-dependent stimulation of glycolysis, but not mitochondrial respiration, by the reproductive neuropeptide GnRH. GnRH stimulation increases gonadotrope GLUT1 expression and translocation to the extracellular membrane. Maximal secretion of the gonadotropin Luteinizing Hormone is supported by GLUT1 expression and activity, and GnRH-induced glycolysis is recapitulated in primary gonadotropes. GLUT1 expression increases in vivo during the GnRH-induced ovulatory LH surge and correlates with GnRHR. We conclude that the gonadotropes of the anterior pituitary sense glucose availability and integrate this status with input from the hypothalamus via GnRH receptor signaling to regulate reproductive hormone synthesis and secretion.Bulk tissue DNA methylation profiling has been used to examine epigenetic mechanisms and biomarkers of complex diseases such as cancer. However, heterogeneity of cellular content in tissues complicates result interpretation and utility. In silico deconvolution of cellular fractions from bulk tissue data offers a fast and inexpensive alternative to experimentally measuring such fractions. In this study, we report the design, implementation, and benchmarking of MethylResolver, a Least Trimmed Squares regression-based method for inferring leukocyte subset fractions from methylation profiles of tumor admixtures. Compared to previous approaches MethylResolver is more accurate as unknown cellular content in the mixture increases and is able to resolve tumor purity-scaled immune cell-type fractions without a cancer-specific signature. We also present a pan-cancer deconvolution of TCGA, recapitulating that high eosinophil fraction predicts improved cervical carcinoma survival and identifying elevated B cell fraction as a previously unreported predictor of poor survival for papillary renal cell carcinoma.
