We aimed to provide a comprehensive aortic stiffness description using magnetic resonance imaging (MRI) in patients with ascending thoracic aorta aneurysm and tricuspid (TAV-ATAA) or bicuspid (BAV) aortic valve.
This case-control study included 18 TAV-ATAA and 19 BAV patients, with no aortic valve stenosis/severe regurgitation, who were 11 age-, gender- and central blood pressures (BP)-matched to healthy volunteers. Each underwent simultaneous aortic MRI and BP measurements. 3D anatomical MRI provided aortic diameters. Stiffness indices included regional ascending (AA) and descending (DA) aorta pulse wave velocity (PWV) from 4D flow MRI; local AA and DA strain, distensibility and theoretical Bramwell-Hill (BH) model-based PWV, as well as regional arch PWV from 2D flow MRI.
Patient groups had significantly higher maximal AA diameter (median[interquartile range], TAV-ATAA 47.5[42.0-51.3]mm, BAV 45.0[41.0-47.0]mm) than their respective controls (29.1[26.8-31.8] and 28.1[26.0-32.0]mm, p<0.0001), while BPces which might help better discriminate such diseases.Multidrug resistant bacterial infections are difficult to treat and contribute to high morbidity and mortality. The phage vB PaeP-SaPL was isolated from a sewage drain (Lahore, Pakistan) against Pseudomonas aeruginosa PA-1 (NCBI Accession number MG763232). Epigenetic inhibitor libraries SaPL produced circular, transparent plaques, 4-5 mm in diameter and showed broad host range infecting 57 % of tested MDR P. aeruginosa clinical isolates (N = 38), while no infectivity was observed against any tested strains of other genera. SaPL inhibited PA-1 growth until 24 h post infection at MOI of 1. The SaPL showed stability at varying temperature and pH, with optimum stability at pH 7 and 45 °C. The latent period of SaPL was 20 min with burst size of 155 virions. The genome of SaPL was double stranded DNA of 45,796 bps having 63 CDS (13 for known proteins and 50 for hypothetical proteins) with a GC content of 52 %. The termini analysis revealed that SaPL genome ends are redundant and permuted. The packaging strategy used by SaPL was a headful (pac) strategy like P1 phage. Survivability of PA-1 challenged mice, treated with SaPL (100 %) was statistically significant (P less then 0.05) than in untreated challenged mice (0%). Based on its efficacy in reducing bacterial growth, selective infectivity against majority of P. aeruginosa strains and its ability to increase survivability in PA-1 challenged mice, SaPL is proposed to be a potential candidate for bacteriophage therapy against difficult to treat MDR P. aeruginosa infections.The resistance of hepatitis C virus (HCV) to direct-acting antiviral agents, used in chronic hepatitis C treatment, consists of a natural process resulting from resistance-associated substitutions (RASs) at specific amino acid regions. To identify and establish the natural prevalence of RASs in the NS3 gene in patients with chronic hepatitis C in the state of Pará, northern Brazil. Molecular analysis was performed on a total of 35 patients infected with HCV genotype 1, who were treatment-naive to protease inhibitors. HCV RNA was extracted from plasma and the NS3 region was amplified and submitted to DNA sequencing (Sanger). The general natural prevalence of RASs in the NS3 gene was 37.5 % (Y56F and S122T). The substitutions Y56F (34.3 %), S122T (3.1 %), V132I (15.6 %) and V170I (9.3 %) were identified. Y56F and S122T provide resistance to the protease inhibitors grazoprevir and simeprevir, respectively. All amino acid substitutions in the NS3 gene, including RASs, identified in patients from the state of Pará were present in other Brazilian studies. The natural presence of RASs in this study reflects the elevated genetic variability of HCV.Action observation and imitation have been found to influence movement in people with Parkinson’s disease (PD), but simple visual stimuli can also guide their movement. To investigate whether action observation may provide a more effective stimulus than other visual cues, the present study examined the effects of observing human pointing movements and simple visual stimuli on hand kinematics and eye movements in people with mild to moderate PD and age-matched controls. In Experiment 1, participants observed videos of movement sequences between horizontal positions, depicted by a simple cue with or without a moving human hand, then imitated the sequence either without further visual input (consecutive task) or while watching the video again (concurrent task). Modulation of movement duration, in accordance with changes in the observed stimulus, increased when the simple cue was accompanied by the hand and in the concurrent task, whereas modulation of horizontal amplitude was greater with the simple cue alone and in the consecutive task. Experiment 2 compared imitation of kinematically-matched dynamic biological (human hand) and non-biological (shape) stimuli, which moved with a high or low vertical trajectory. Both groups exhibited greater modulation for the hand than the shape, and differences in eye movements suggested closer tracking of the hand. Despite producing slower and smaller movements overall, the PD group showed a similar pattern of imitation to controls across tasks and conditions. The findings demonstrate that observing human action influences aspects of movement such as duration or trajectory more strongly than non-biological stimuli, particularly during concurrent imitation.
