B cells are a critical arm of the adaptive immune system. After encounter with antigen, B cells are activated and differentiate into plasmablasts (PBs) and plasma cells (PCs). Although their frequency is low, PB/PCs can be found in all lymphoid organs including peripheral lymph nodes and spleen. Upon immunization, depending on the location of where B cells encounter their antigen, PB/PCs subsequently home to and accumuate in the bone marrow and the intestine where they can survive as long-lived plasma cells for years, continually producing antibody. Recent evidence has shown that, in addition to producing antibodies, PB/PCs can also produce cytokines such as IL-17, IL-10, and IL-35. In addition, PB/PCs that produce IL-10 have been shown to play a regulatory role during experimental autoimmune encephalomyelitis, an animal model of neuroinflammation. The purpose of this review is to describe the phenotype and function of regulatory PB/PCs in the context of experimental autoimmune encephalomyelitis and in patients with multiple sclerosis.Obesity and anxiety are public health problems that have no effective cure. Obesity-induced anxiety is also the most common behavioural trait exhibited amongst obese patients, with the mechanisms linking these disorders being poorly understood. AP20187 and hippocampus are reciprocally connected, important neurogenic brain regions that could be vital to understanding these disorders. Dietary, physical activity and lifestyle interventions have been shown to be able to enhance neurogenesis within the hippocampus, while the effects thereof within the hypothalamus is yet to be ascertained. This review describes hypothalamic neurogenesis, its impairment in obesity as well as the effect of interventional therapies. Obesity is characterized by a neurogenic shift towards neuropeptide Y neurons, promoting appetite and weight gain. While, nutraceuticals and exercise promote proopiomelanocortin neuron proliferation, causing diminished appetite and reduced weight gain. Through the furthered development of multimodal approaches targeting both these brain regions could hold an even greater therapeutic potential.Neurodegenerative disorders are common among aging populations around the globe. #link# Most are characterized by loss of neurons, protein aggregates, oxidative stress, mitochondrial damage, neuroinflammation among others. Although symptomatic treatment using conventional pharmacotherapy has been widely employed, their therapeutic success is limited due to varied reasons. In the need to identify an alternative approach, researchers successfully demonstrated the therapeutic utility of plant-derived nutraceuticals in cell and animal models of neurodegenerative conditions. However, most nutraceuticals failed during clinical trials in humans owing to their poor bioavailability in vivo and limited permeability across the blood brain barrier (BBB). The current emphasis is therefore on the improved delivery of nutraceuticals to the brain. In this regard, development of nanoparticle conjugated nutraceuticals to enhance bioavailability and therapeutic efficacy in the brain has gained attention. Here, we review the research advances in nanoparticles conjugated nutraceuticals applied in neurodegenerative disorders and discuss their advantages and limitations, clinical trials and toxicity concerns.Bacterial cellulose (BC), prepared from two recently developed thermotolerant bacterial strains (Komagataeibacter xylinus C30 and Komagataeibacter oboediens R37-9), were used as a raw material to synthesize nanofibril films. Field-emission scanning electron microscope (FE-SEM) observations confirmed the ultrafine nano-structure of BC pellicle (BCP) with average fibril widths between 50 and 60 nm. The BC was directly oxidized in a TEMPO/NaBr/NaClO system at pH of 10 for 2 h. TEMPO-oxidized bacterial cellulose nanofibrils (TOBCN) were obtained by a mild mechanical treatment and the TOBCN films were prepared through heat-drying. The oxidation yielded a recovery ratio between 70 and 80% by weight with an increase in the carboxylate content of 0.9-1.0 mmol g -1. Nanofibrillation yields were more than 90% and the resulting high aspect ratio TOBCNs were ~6 nm in average width with >800 nm in lengths, when observed under transmission electron microscope (TEM). TOBCN film of K. xylinus C30 exhibited high transparency (79%), tensile strength (142 MPa), Young’s modulus (7.13 GPa), elongation around failure (3.89%), and work of fracture (2.29 MJ m-3), when compared to the TOBCN films of K. oboediens R37-9 at 23 °C and 50% RH. Coefficients of thermal expansion of both the TOBCN films were low at around 6 ppm K-1.A facile route towards new chitin derivatives with both catalytical and biological activities is proposed in the course of methodology development aimed at the design of polyfunctional materials on the basis of renewable and accessible natural polysaccharides. Ultrasound-promoted and Cu(I)-catalyzed azido-alkyne click cycloaddition of the propargylic ester of nicotinic acid and its N-methylated analogue to the azido chitin derivative allowed us to obtain previously unknown non-toxic water-soluble derivatives of chitin. The obtained polymers and their based nanoparticles demonstrated a high antibacterial activity in vitro, which is comparable or even superior to that of commercial antibiotics ampicillin and gentamicin. New derivatives of chitin were also shown to be highly efficient and reusable (at least for 10 times) green catalysts for the aldol reaction in water. The catalysts can be easily separated from the reaction mixture by their precipitation with ethanol. The results obtained highlight prospects of further studies on chitin’s application in the rational design of novel functional materials with valuable properties.Rebaudioside D is a promising sweetener due to its zero calorie and high sweetness. Here, a transglucosylase gene eugt11 from Oryza sativa was for the first time expressed in Pichia pastoris, and transformant XE-3 showed the highest expression levels in pH 5.5 BMMY media containing 0.75% methanol. The affinity-purified EUGT11 from XE-3 displayed the highest activity at pH 6.0-6.5 and 45 °C, compared to pH 8.5 and 35 °C for EUGT11 from Escherichia coli. One-pot synthesis with orthogonal design was employed to optimize the rebaudioside D production using XE-3, and the initial pH 7.0 of the medium appears to be a significant factor and delivers the highest conversion efficiency. A two-step temperature-control strategy was developed, and a conversion rate of 95.31% was achieved at 28/35 °C vs. 62.41% in a one-step process at 28 °C. This study provides a high-efficient whole-cell biocatalysts technology for the sweetener production.
