However, it is still unknown whether silencing Pellino1 (si-Pellino1) has a therapeutic effect on this disease. Here, we showed that silencing Pellino1 can be a potential protective strategy for abnormal myocardial energy metabolism in LPS-induced myocarditis. We used liquid chromatography electrospray-ionization tandem mass spectrometry (LC-MS/MS) to analyze samples from si-Pellino1 neonatal rat cardiac myocytes (NRCMs) treated with LPS or left untreated. After normalization of the data, metabolite interaction analysis of matched KEGG pathway associations following si-Pellino1 treatment was applied, accompanied by interaction analysis of gene and metabolite associations after this treatment. Moreover, we used western blot (WB) and polymerase chain reaction (PCR) analyses to determine the expression of genes involved in regulating cardiac energy and energy metabolism in different groups. LC-MS-based metabolic profiling analysis demonstrated that si-Pellino1 treatment could alleviate or even reverse LPS-induced cellular damage by altering cardiomyocytes energy metabolism accompanied by changes in key genes (Cs, Cpt2, and Acadm) and metabolites (3-oxoocotanoyl-CoA, hydroxypyruvic acid, lauroyl-CoA, and NADPH) in NRCMs. Overall, our study unveiled the promising cardioprotective effect of silencing Pellino1 in LPS-induced myocarditis through fuel and energy metabolic regulation, which can also serve as biomarkers for this disease.In the summer season, the harmful effects of high ambient temperatures on rabbit productivity have attained global interest. Therefore, the use of new nutritional strategies to improve heat tolerance in rabbits has become highly needed. In the current experiment, the effect of the addition of L-carnitine (LC) to normal (NE) or high-energy (HE) diets of growing rabbits reared under summer conditions on their productive performance, physiological indicators, and carcass characteristics was assessed. For this purpose, a 2 × 2 factorial arrangement of treatments was used with two dietary levels of energy of 10.00 and 11.22 MJ kg-1 diet of NE and HE, respectively, which was fortified with 0 or 50 mg of LC kg-1 of diet (LC0 and LC50, respectively). The feeding trial was conducted using 80 male rabbits (initial body weight, 630.7 ± 1.3 g; 35-day-old), and it lasted for 56 days. Throughout the experiment, the average temperature humidity index was 35.49 reflecting very severe heat stress conditions. Rectal temperature, heart rate, and respiration rate decreased significantly in rabbits fed NE+LC50, HE+LC0, and HE+LC50 diets compared with those fed NE diets without supplementation. Simultaneously, growth indices and feed conversion ratio were enhanced significantly. Rabbits fed NE+LC50, HE+LC0, and HE+LC50 diets showed significantly higher blood hemoglobin, white blood cell counts, total protein, glucose, and red blood cell counts, compared with those fed a NE+LC0 diet. Rabbits fed NE+LC50 and HE+LC50 showed higher economic efficiency. In conclusion, growing rabbits reared under summer conditions responded positively to LC supplementation and HE diets, in terms of the highest growth performance, feed conversion, and physiological and biochemical indicators, making them potentially safe to use.

    Studies indicate that other cardiovascular problems than aortic disease are a burden for patients with Marfan syndrome (MFS). The aim of the study was to assess the extent of this issue.

    A registry-based population study of patients with a Ghent II verified MFS diagnosis. Each patient was matched with up to 100 controls on age and sex. From the Danish healthcare system, we identified 407 MFS patients (from 1977 to 2014) and their cardiovascular events and compared them with those in 40,700 controls. Total follow-up time was 16,439 person years.

    Mitral valve disease was significantly more common in MFS [HR 58.9 (CI 38.1-91.1)] and happened earlier and more often in women than men with MFS [age at first registration 22 vs. 38years, HR 2.1 (CI 1.0-4.4)]. Heart failure/cardiomyopathy was also more common in MFS [HR 8.7 (CI 5.7-13.4)] and men were more affected than women, and at younger age [39 vs. 64years, HR 0.18 (CI 0.06-0.55)].
