Our results provide a new link between ZIC2 and human cardiogenesis and document the potential power of a genome-wide unbiased CRISPR-knockout screen to identify the key steps in human mesoderm precursor cell- and heart progenitor cell-fate determination during in vitro hPSC cardiogenesis. ©AlphaMed Press 2020.Perovskite single crystals (PSCs) possess superior optoelectronic properties compared to their corresponding polycrystalline films, but their applications of PSCs in high-performance, integrated devices are hindered by their heavy thickness and difficulty in scalable deposition. Here, a microchannel-confined crystallization (MCC) strategy to grow uniform and large-area PSC arrays for integrated device applications is reported. Benefiting from the confinement effect of the microchannels, solution flow dynamics is well controlled, and thus uniform deposition of PSC arrays with suitable thickness is achieved, meaning they are applicable for scale-up device applications. The resulting PSCs possess excellent optoelectronic properties in terms of a long carrier lifetime (175 ns) and an ultralow defect density (2 × 109 cm-3 ), which are comparable to the corresponding bulk crystals. The unique embedded structure of PSCs within the microchannels allows the construction of a high-integration image sensor. KGaA, Weinheim.With an estimated 425 million diabetic patients worldwide in 2019, type 2 diabetes (T2D) has reached a pandemic proportion and represents a major unmet medical need. A key determinant of the development and progression of T2D is pancreatic beta-cell dysfunction, including the loss of cell mass, the impairment of insulin biosynthesis, and inadequate exocytosis. Recent studies have shown that transient receptor potential vanilloid 4 (TRPV4), a Ca2+ -permeable nonselective cation channel, is involved in beta-cell replication, insulin production and secretion. TRPV4 agonists have insulinotropic activity in pancreatic beta-cell lines, but the prolonged activation of TRPV4 leads to beta-cell dysfunction and death. In addition, TRPV4 is involved in a wide variety of pathophysiological activities and has been reported to play an important role in diabetes-related complications such as obesity, cardiovascular diseases, diabetic retinopathy, nephropathy, and neuropathy. In a rodent T2D model, Trpv4 agonists promote vasodilation and improve cardiovascular function, while Trpv4 antagonists reduce high-fat diet-induced obesity, insulin resistance, diabetic nephropathy, retinopathy, and neuropathy. These findings raise interest in using TRPV4 as a therapeutic target for T2D. In this review, we intend to summarize the latest findings regarding the role of TRPV4 in diabetes as well as diabetes-related conditions and to evaluate its potential as a therapeutic target for diabetes and diabetes-related diseases. All rights reserved.BACKGROUND Metastatic head and neck cancers (HNCs) predominantly affect the lungs and have a two-year overall survival (OS) of 15% to 50%, if amenable for pulmonary metastasectomy. METHODS Retrospective review of the two-year local control (LC), local-regional control (LRC) within the same lobe, OS, and toxicity rates in consecutive patients with metastatic pulmonary HNC who underwent stereotactic ablative radiotherapy (SABR) January 2007 to May 2018. RESULTS Evaluated 82 patients with 107 lung lesions, most commonly squamous cell carcinoma (SCC; 64%). Median follow-up was 20 months (range 9.0-97.6). Systemic therapy administered in 34%. LC, LRC, and OS rates were 94%, 90%, and 62%. Patients with oligometastatic disease had a higher OS than polymetastatic disease, 72% vs 44% (HR = 0.30, 95% CI 0.14-0.64; P = .008). OS in oligometastatic non-SCC and SCC were 100% and 66% (P = .03). There were no grade ≥3 toxicities. CONCLUSIONS Metastatic pulmonary HNCs after SABR have a two-year OS rate comparable to pulmonary metastasectomy. © 2020 Wiley Periodicals, Inc.The effect of preoperative external radiotherapy (XRT) on head and neck free-flap reconstruction is still controversial. In this study, we aimed to determine the impact of preoperative XRT on the outcomes of head and neck microvascular free-flap reconstruction. ML792 A systematic review and meta-analysis was conducted in concordance with the Systematic reviews and Meta-Analyses protocol. We searched several databases (PubMed, EMBASE, Web of Science, and Cochrane Library) to find published papers on the topic. The R program was used for data synthesis and statistical heterogeneity evaluation; then, fixed effect or random effect models were used. A total of 37 studies, involving 12 408 patients with 12 668 flaps, were included in this meta-analysis. The overall flap success rate for all studies was 94.4%. Pooled analysis showed that patients with preoperative XRT were significantly associated with an increased risk of total flap failure (odds ratio [OR] = 1.80, 95% confidence interval [CI] = 1.45-2.23, P less then .001), partial flap failure (OR = 1.90, 95% CI = 1.07-3.38, P = .029), and postoperative complication rates (OR = 2.22, 95% CI = 1.64-2.72, P less then .001). Our study suggests that preoperative XRT is associated with an increased risk of developing free-flap failure and an increased postoperative complication rate. © 2020 Wiley Periodicals, Inc.The electron-transport-layer (ETL) free perovskite solar cells (PSCs) are attractive because of fewer layers and hence lower cost, but the lower photovoltaic performance, as compared to the ETL-contained PSCs, largely restricts their practical applications. Herein, we design and synthesize hydroxylethyl functionalized imidazolium iodide, whose single crystal structure is determined, and propose self-assembled ionic liquid on the conductive substrate for ETL-free PSCs. It is found that the self-assembly of the ionic liquid on the conductive substrate can lower the work function of the conductive substrate, enhance the interfacial electron extraction, and meanwhile retard the interfacial charge recombination. As a consequence, the power conversion efficiency is improved remarkably from 9.01% to 17.31% upon the self-assembly of ionic liquid on the conductive substrate.
